Metabolic Dysfunction-Associated Steatohepatitis is liver inflammation and damage caused by buildup of liver fat, often associated with obesity, T2D, dyslipidemia, and hypertension. MASH may progress to fibrosis and cirrhosis, increasing the risk of liver-related morbidity and mortality, and while disease progression generally is slow, some patients rapidly progress to advanced fibrosis. As no treatments are yet approved for MASH, interest in development of therapeutic options for non-cirrhotic and cirrhotic MASH is high. Drug development is challenged by heterogeneous disease etiology and pathophysiology, slow disease progression requiring surrogate markers to predict effect on clinical outcomes, and dependence on biopsy for diagnosis which has spurred extensive research into non-invasive tests. While multiple drug classes show promise to improve MASH and/or fibrosis, combination therapy may be needed to ameliorate disease severity or halt progression.
